Prepare Your Workspace

  • Wash your hands and wear clean clothes.


  • Close any windows and doors to reduce airflow and minimise the risk of airborne contaminants.

  • Wipe down the worktop with 70% isopropyl alcohol or a bleach-based surface cleaner to disinfect.

  • Use the included gloves and alcohol wipes to keep everything sterile.

Inoculate the Grain Bag

  • Carefully attach the sterile needle to your spore or liquid culture syringe without touching the needle or removing the cap.

  • Wipe injection port and surouding area with a 70% alcohol wipe provided

  • Remove the needle cover and inject 1–2 ml of spores or liquid culture into the grain bag.

    Note: Spores often clump together. Please shake the syringe thoroughly to break them up before injecting.


  • Gently shake the bag to help spread the inoculant evenly through the grain.

  • Place the bag in a dark environment at 20–25°C to allow the mycelium to grow.

    Important: Do not use heat mats - they can overheat and kill the mycelium. Instead, heat the space, not the bag.

Colonise the Grain

  • Once your grain is 20–30% colonised, it’s time to Break & Shake the mycelium and redistribute it evenly throughout the bag.

    This step is optional, but it can significantly speed up the colonisation process.

  • After shaking, place the bag back into incubation and wait until it’s fully colonised.

  • Important: Please ensure your grain is fully colonised before moving to the next stage.

    We recommend leaving it for a few extra days, even after it appears ready, to give the mycelium time to consolidate.

    Exposing uncolonised grain is the number one cause of contamination during the next step.

Mix the Grain with Substrate

  • Once your grain is fully colonised, it’s time to mix it with the substrate.

    Gently break the mycelium apart into loose, individual grains while still inside the grain bag.


  • Cut the tops off both the grain and substrate bags

  • Pour the broken-up grain into the substrate bag. Fold the top of the substrate bag a few times and gently mix the contents to distribute the grain evenly throughout the substrate.

  • Use the paperclips provided to secure the bag closed.rubber band.
  1. Incubate againPlace the bag back in your incubation area and allow it to fully colonise under the same conditions used for grain — ideally 20–25°C in darkness.
